The PhD will be attached to the Doctoral School of Chemical Sciences of the University of Strasbourg (ED222).
The PhD student will join the LSMBO team of the IPHC and will be able to benefit from all the material conditions (LC-MS / MS instrumentation), human support (sharing of expertise) and IT (access to all the software tools for the processing of LC-MS / MS data) necessary for the successful completion of this thesis project.
The thesis will take place in the context of a European H2020 Marie Sklodowska Curie Actions (MSCA ITN 2019) research program (TACT Targeted Anti-Cancer Therapy project). Thus, the PhD student will have to collaborate / exchange results with all the partners of this international consortium. Also secondaments in other labs are foreseen.

Thesis subject : New MS-based approaches for Protein Drug Conjugate (PDC) characterization at intact & middle- levels
Objectives of the thesis : Monoclonal antibodies (mAbs) and their related compounds make up the largest class in human therapeutics to treat many diseases. Their success stems from their high specificity and affinity, long circulating half-lives, ability to induce immune cell effector response, and structural versatility. However, the analytical characterization of mAb-based formats is challenging due to many sources of heterogeneities related to charge and size variants (multimers and aggregates) and primary sequences. As narrow acceptance criteria, defined as Critical Quality Attributes (CQA), are requested by regulatory agencies (FDA and EMA), suitable analytical techniques are required to analyze those heterogeneities qualitatively and quantitatively. In particular, additional specific criteria are requested for PDCs, for which bioconjugation brings an additional level of heterogeneity. State-of-the-art chromatographic and mass spectrometric methods lay at the forefront of mAb-format characterization, due to their complementarity and orthogonality. The analytical characterization of mAb-related compounds usually follows a multi-level workflow: intact protein level (top level), smaller sub-domains analysis after limited enzymatic digestion (middle level), and peptide level upon combination of several proteases (bottom level). In the latter case, the extended enzymatic digestion can affect the quality of the final product leading to the loss of key features. Top- and middle-level are performed without any sample preparation, or a very reduced digestion step, which make these approaches of utmost interest, but are still in their infancy and often performed in denaturing conditions, leading to the loss of the active conformation of the mAb-format. In that context, this PhD project will be at the interface of analytics, biochemistry and biology, and aims to develop new intact and middle-level MS-based approaches for PDC characterization.

Additional comments

The candidate must hold a master's (MSc) degree.
He/she will need to prove a solid knowledge of protein biochemistry, in mass spectrometry and chomatography. Experience in native MS or top dawn MS will be a string advantage.
The candidate will have to master English fluently in writing and orally in order to be able to write results reports and present them at meetings of the TACT consortium.
The international, multidisciplinary and collaborative context of this project requires that the candidate has a strong motivation, curiosity to expand his area of expertise, autonomy and ability to work in a team with strong constraints to meet deadlines.
